Synapse degeneration is held to be an early and critical pathophysiological event in Alzheimer's disease; however, the mechanisms involved are not understood. Here we aim to analyze the kinetics of dendritic spine loss throughout disease progression in mouse models of AD by applying long-term 2-photon in vivo imaging. Using different transgenic animal models of familial Alzheimer's disease with no Aβ-pathology, Aβ-pathology only or both Aβ- and tau-pathology, as well as lentiviral transfection of single cortical neurons which modifies APP expression, and proteases involved in Aβ generation or degradation, we aim to understand if and how Aβ alters dendritic spines. Moreover, we are going to analyze the effects of therapeutic approaches that have been mechanistically associated with spine loss in AD, namely a NMDA-receptor open-channel blocker and inhibitors or modulators of the γ-secretase and β-secretase.
